The accident occurred shortly before 1 p.m.
Police said an Oakville man in his 40s, who was driving a Grey Compact, was found with vital signs absent.
He was transported to Oakville Trafalgar Memorial Hospital where he was pronounced dead.
A woman was also taken to hospital with non-life threatening injuries.
Det. Const. Chris Heffernan said the Halton Regional Police Service Collision Reconstruction Unit is investigating the possibility that a medical episode may be behind the collision.
The crash initially closed both lanes of Third Line, however, they have since reopened.
